Optimization of ¥â-1,3/1,6-Glucan Production of Aureobasidium pullulans KSY-0516 Strain

Abstract
¥â-Glucans are major bioactive compounds known to have biological activities, including anti-cancer, anti-inflammatory, and immune-modulating properties. Aureobasidium pullulans, known as black yeast, produces a variety of black ¥â-glucans, which can be used to produce white ¥â-1,3/1,6-glucan using a mutation technique. The purpose of this study was to determine the optimum production yield of ¥â-1,3/1,6-glucan using response surface methodology, which is a statistical procedure used for optimization studies. A central composite design was applied to investigate the effects of independent variables, sucrose concentration (X1), KNO3 concentration (X2), and oxalic acid concentration (X3) on responses such as yield and ¥â-1,3/1,6-glucan production rate. A maximum yield of 1.86217 g/L was obtained at 4.74% sucrose, 0.19% KNO3, and 0.036% oxalic acid. Based on the above results, the optimal production and efficient production of ¥â-glucans can be achieved.
